Concentration and Quality Control
This service is only for future sequencing projects at CTG.
Requesting a project at CTG
To initiate a project using our available services, please start a project in iLab. You can login using your LucatID (SWAMID) on the Lund University iLab webpage.
RNA concentration
RNA Concentration is measured by a fluorometric method, using the Qubit 4 or Qubit Flex instruments.
Available Assays:
- Broad Range RNA Assay
- High Sensitivity RNA Assay
RNA quality
RNA quality is assessed by micro-capillary gel electrophoresis, using the TapeStation 4200. RNA sample quality is expressed as RNA Integrity Number (RIN) that can vary from 1 to 10, with the higher number indicating higher quality.
Available assays:
- RNA Assay
- High Sensitivity RNA Assay
Sample Requirements
Detailed instructions will be provided after requesting a QC project in iLab.
Deliverables
Concentration measurement results will be delivered as:
- Excel file containing the measured values.
Quality measurement results will be delivered as:
- PDF files containing electropherogram and RIN values.
- Excel file containing RIN values.
DNA concentration
DNA Concentration is measured by a fluorometric method, using the Qubit 4 or Qubit Flex instruments.
Available Assays:
- Broad Range dsDNA Assay
- High Sensitivity dsDNA Assay
DNA Quality
DNA quality is assessed by micro-capillary gel electrophoresis, using the TapeStation 4200. DNA sample quality is expressed as DNA Integrity Number (RIN) that can vary from 1 to 10, with the higher number indicating higher quality.
Available assays:
- Genomic DNA Assay
- Cell-free DNA Assay
Sample Requirements
Detailed instructions will be provided after requesting a QC project in iLab.
Deliverables
Concentration measurement results will be delivered as:
- Excel file containing the measured values.
Quality measurement results will be delivered as:
- PDF files containing electropherogram and DIN values.
- Excel file containing DIN values.
DNA Quality for FFPE samples
In addition to concentration measurements described for DNA above, a qPCR is performed to assess the quality of FFPET-derived DNA more accurately. This assay gives an estimation of DNA fragmentation status and its amplification potential expressed as ∆Cq value.
Sample requirements
- Sample concentration needs to be determined by a fluorometric method (e.g. Qubit).
- Sample needs to be normalized in EB-buffer (10 mM Tris-Cl, pH 8.5).
- Concentration: 2.5 ng/µl*
- Volume: 5 µl
* Samples with concentrations lower than 2.5 ng/µl cannot be processed
- Samples should be delivered in 200 µl strips.
- All samples should be labeled individually, directly on the side and the lid of a tube.
- Samples must be delivered on ice.
Deliverables
Quality measurement results for FFPE samples will be delivered as:
- Excel file containing ∆Cq scores and the quality estimates.
