Sample Requirement

DNA Extraction

  • We recommend Qiagen spin column-based nucleic acids purification methods.

 Concentration and Purity

  • Use spectrophotometrical method to get the concentration and a ratio (OD260/280 and OD260/230) of each sample.
  • DNA should have an OD260/280 ratio ~1.8 and OD260/230 ratio in range 1.8-2.2 (the ratio shows purity of samples).

DNA amount recommended

  • Methylation: 
    • Bisulfitconversion: 555 ng DNA in 50 uL nuclease-free ultrapure H2O (11.1 ng/μL).
       
  •  SNP genotyping, CNV: 600 ng DNA in 10 μL nuclease-free ultrapure H2O (60 ng/μL). Lower amounts can be discussed however the minimum volume is usually 4 μl.

  • Order Form and Sample sheet (provided by CTG) have to be filled in and sent to facility by email (CTGservice [at] med [dot] lu [dot] se (CTGservice[at]med[dot]lu[dot]se)).
  • Use Checklist for DNA delivery (provided by CTG)
